上个长周末,忙完烦索的家务事,早上5：30我怀着急切的心情来到了当地附近的一个水
库.由于我的目标鱼是皮皮,所以只带了个ultra light的杆,绑了6lbs的线和1/16oz的
jig head钩子,用的是grub的软饵,手边也连个魚网抄子也没带,就在岸边甩杆子开钓了.
现在正值晚春,水库近岸水草很多,钩子动不动就挂上了水草. 也许是因为温度上升太
快,或是时节原因皮皮甩籽产卵已完毕,钓了近半个小时,只上了一条6”的皮皮,因为尺
寸不够(我们这儿皮皮keep的尺寸是9”above)就给放了. 正在士气低沉,甩杆出去往回
收时,就觉得钩子很重,以为又挂到草了,于是就大力往回拉. 这一拉不要紧,就觉得眼前
水中魚影一翻,好家伙,好大的一条呀！(至少对于我来说这鱼不小,因为我脑子里一直想
着的是Crappie的印象). 真是心头一惊,菊花一紧,人立即进入战斗状态,身体里肾上腺
素急剧上升.我立即把我的UL杆子头往上抬紧,边调整轮子的阻尼. 耳边随即响起了轮子

实验室需要做Northern，但之前没人做过。现在有500-1000bp的被Digoxigenin标记的
Anti-sense RNA探针。有Li-cor Odyssey的远红外检测的仪器。现在想找合适的试剂设
计一下怎么做，和大家讨论讨论。
方案1. 看了一下Roche的那个DIG NORTHERN STARTER KIT，最后用的是anti-
digoxigenin-AP。查了一下Odyssey的Fluorescent Reagent Compatibility，Odyssey
能检测NBT/BCIP，但是可能不能定量。
方案2. 抗体直接找一个Anti--digoxigenin-X，X能被Odyssey检测，比如IRdye，Alexa
680什么的。
方案3. 先找个一抗比如mouse/rabbit anti-Digoxigenin，然后用已有的二抗anti-
mouse/rabbit-IRdye680/800CW来检测，有点像Western blot。

我现在怀孕21周，在生化试验室工作。自己没有做过任何放射性试验，但试验室里有很
多人做。怀孕后我都倍加小心，但是今天竟然疏忽了！在一个oven边上前后累计操作试
验一小时左右，之后才发现oven里面有人正在做northern blot，而且中间还拿放
northern blot的玻璃试管两次！
发现以后真实后悔不迭。不知道这在oven前面放射性还大不，会对肚子里的宝宝有影响

很多（total RNA和polyA RNA都是），真正的条带相对很微弱。可是奇怪的用这个
probe做的FISH的结果很干净，信号看着一点不像backgroud. target出现在部分neuron
里，non-neuron里完全没有。
想请教试验大牛们，这FISH结果能相信吗？
如果FISH能crosshyb到rRNA上，看上去啥样？
怎样能改善northern的结果呢？polyA RNA仍然不能完全清除rRNA,所以还是有crosshyb
.
