function）作为积木块，拼积木拼出巨大 piece-wise function，加 function of
function。这样 function 可以很复杂，内在参数无数，变化无穷，才可以上大数据。
其实就好比把 iterative numerical solver 的思想用在统计学 regression fitting
上。当然这个比方不是很确切。
: 以前这些方法不好使，是因为数据量不够大。要消除不确定性或者噪音就

也不容易稳定。
我觉得工业界里 iterative numerical solver 写 costing 就是个 arts 而不是
science，convergence 好不好主要看手气。
深神经网络的 layered network 咋玩我不懂，但搞不好也是个艺术活。

了耳鼻喉科医生，医生建议做个脑部的MRI.做完后医生看过了报告后说脑部没什么异常
。 不知道有没有必要再找脑神经科的医生看一下？ 报告如下。小时侯得过鼻窦炎，出
国后就没什么症状了。平时有时候会有头疼，主要集中在脑门部位。喝杯咖啡，或者睡
一觉就没事了。

刚开始学，感觉black box效果不会好，需要经验和优化， 比如说
1. sigmoid （logistic） function 用在股价数据（连续）是否合适？
2. 怎么搭architecture效果更好
3. 怎么tune parameters, 要不要weight decay?
建好模型，data 倒是好多，历史数据， 相关股票的信息，把政治因素突发事件考虑进
去都有可能 （以前也加过息啊，竞选过总统啊，把那段时间的数据拿来learn一下不就
行了） |

工业上。本身就是数字滤波器往前一小步。
以前是成本低，懂的人少。
这波泡沫过后，成本就下来了。
我觉得只要会的人多
用的就会多一些

哎....
1.
早先神经网络一个人叫罗什么。和明斯基是高中同学。
罗什么是最早一种神经网络的鼓吹人和计算机程序实现人。
名噪一时。据说得罪了不少人。明斯基早先是他这边的。
后来明斯基写了本名著攻击神经网络。这明星学者罗什么
早早的就死了。不知是否自杀。

然而，当时有个不是那么出名的分类器，叫做
widrow-hoff学习算法什么的。widrow在这摊子烂事
出来之后，就不提神经网络了。本身widrow就是EE的。
也不和这帮人一个资金来源。

为了实现硬件方便，
Widrow 把Sigmoid改成y=x纯线性的滤波器，只保存
学习办法，也就是approximate gradient.还有网络结构，
就是线性权值互联。这就是世界上应用最广泛的least mean square 滤波器的来源。
今日我们很难想象，数字滤波器的一大源头其实是神经网络。但在当时是很自然的。因
为那时候还没有大规模的
大规模的线性数字滤波器件。
后来社会需要大规模集成电路上的各种自适应滤波--也就是学习。不然自适应个蛋啊。

发帖数: 320 | 36 Real-time PCR 的Ct 本身是特指刚能够测到信号时，需要的 cycle 数。这个cycles
数 和原始样本的量的“对数”成线性关系。当你调节 threshold 的时候，离本底越远
，也就离线性关系越远。到最后大家都饱和了，就和普通 PCR 跑胶一样了。当然，在
理想情况下，“稍微”离开一点，对线性关系的影响应该不大。但这时候，软件找到的
应该是最佳的。
如果反应不理想，比如本底比较强，或者信号上升曲线不理想，最好的方法是把
reaction optimized。实在要自己调的时候，一般是本底杂信号高的时候（左边本来应
该低于 threshold 的信号有时候会超过 threshold），你估计可以稍微调节一下
threshold 到刚好超过背景信号的地方。尽量不要离开太远，否则会导致离开线性关系
太远。
顺便介绍一下第二个方式（F0）：
除了Ct的方式，还有如果上升曲线很好，也可以用一个算 F0 的方式。也就是通过非线
性回归（Sigmoidal）计算样本没有放大的时候的应该有的原始荧光强度（F0 ＝cycle

的标准抗体做了standard curve。然后根据检测到的OD推算抗体浓度 。用的是Prism软
件， 不知道为什么很多样品OD值软件没有给出浓度。 这些样品OD值一般在standard
curve的两极（最高值 或者最低值，或超出standard curve）。curve fit的时候用的
是 sigmoidal, 4PL, X is log[concentration]。请问板上的大牛们， 我应该怎么做
才能得到所有样品OD值 对应的抗体浓度。
