第一篇 DNA测序， Sanger Style - 纪念Frederick Sanger
2013年11月20号，Frederick Sanger 去世了， 享年95岁。Sanger是DNA测序之父，获
得过两次诺贝尔奖， 一个是1958年蛋白测序， 另一个是1980年DNA测序。虽然DNA结构
发现的比蛋白晚， 但毕竟是genetic code，master script。ATCG四个碱基的组合比蛋
白的20个氨基酸的天文数字组合可更容易研究找规律了。人天生就是个好奇的动物，
最喜欢找规律, 尤其喜欢从简单的地方入手，所以围绕DNA的研究在Watson-Crick的碱

看了Leon Avery的宣言“Leaving Science”，真为他感到轻松。虽然不同意他的做法
，但还是佩服他的勇气。如果对一件事情已经失去了兴趣，并总在自我怀疑而又觉得
forever trapped了的时候，再怎么做都会让自己很纠结而痛苦。对于自己和周围的人
都不是件好事，那还不如快刀斩乱麻的放下。感觉他不是“leaving science”, 而是

Spring Harbor）实验室的那次会议已经是一年多前的事情了。但那天发生的事情至今
还历历在目,回忆起来仍觉得有趣。
这是冷泉港纪念“数量生物学”（Quantitative Biology）会议75周年的年会，但会议
的内容已不限于数量生物学，内容囊括了生物学大部分的前沿领域，与会者也是“名人
云集”，当然其中最显眼的莫过于诺贝尔奖得主，DNA双螺旋结构的发现者之一James
Watson了。
会议首先由冷泉港实验室主任Bruce Stillman致欢迎辞。Stillman于1994年从James
Watson手中接任冷泉港实验室主任，并于2003年获得连任，为英国皇家学院（The
Royal Society）、美国国家科学院（National Academy of Sciences）、美国文理学
院（American Academy of Arts and Sciences）院士。做为冷泉港的总裁，他果然有
管理者的风范，举止得体，致辞周到，但让我觉得未免有点俗套。

小看海底两万里的，然后就加入海军，当了commander，也拿了一个PhD，然后也是成了
Professor，其间，发现了除了Titanic，还有Bismarck，然后就是Yorktown，还有JFK
的鱼雷快艇的残骸。
然后，就是那个James Cameron，原来是开卡车的，然后混入电影界，然后就混出头了
，搞搞海底探险，业余也拍两部电影什么的，原来小时候也是看海底两万里的。看来这
个海底两万里确实是Inspiring。
生物，嗯，如果像Craig Venter那样搞条船，周游一下世界，也算是有点那个意思。好
像凡尔纳也有一条豪华yacht，有空也周游一下世界什么的，很爽的。那个Venter的巫
师二号也很豪华的。不过，这样就是海上两万里了。 |

发帖数: 4339 | 34 刚对比了一下venter 2010合成细菌的文章，这项工作在技术上没有什么突破，而且看
他们合成的策略，是在野生型酵母里一段一段（30KB）的更换成合成序列，比起venter
的在酵母里合成好完整的细菌基因组，再转移到细菌细胞质里，看上去难度还要小一些。
但是意义还是很大的，毕竟是真核生物里的第一次。至于有什么实用或者带来什么新知
识，我暂时还没有看到。意义大于内容吧。 |

这个领域的确很火，竞争也很激烈，真正火起来是next-gen sequencing 技术出来以后
，也就是过去15年内。
但他对于单分子酶学的研究室是在20多年以前，算是这个领域的先驱之一。
单分子/单细胞分离分析仪器，这种不是一个人、一个组能做的工程；当年venter发明
鸟枪法也是基于之前在公共研究机构的长期积淀的结果。

同时在线发表了2篇将酵母染色体融合的成果。其中一篇来自酵母染色体人工合成领域
“老将”、美国科学院院士、纽约大学医学院教授杰夫•博克（Jef Boeke）团队
，另一篇则来自中国科学院分子植物科学卓越创新中心/植物生理生态研究所合成生物
学重点实验室覃重军研究团队及其合作者。
中国研究成果的通讯作者之一、植物生理生态研究所研究员、中国科学院院士赵国屏对
澎湃新闻表示，“这是继上世纪六十年代人工合成胰岛素和tRNA之后，中国学者再一次
用合成科学的方法，回答了一个生命科学领域十分基础而重大的问题。”
