我目前用的von Kossa, 但是结果 有问题，请问有人做过von kossa 干细胞
分化染色的，negative 和positive的都是阳性。
这是protocol：
1.ethanol -20 10min
2.wash 3 times with tap water (这里可以用tap water 么？ 它里面应该有钙吧）
实验员说不能用distilled water, 因为钙会溶解在里面？？
3. 3% Silver Nitrate 30 mins RT 没有特别用光照，就放在实验室里，上面
有日光灯
4. 3 wash tap water
5. 碳酸钙 福尔马林 2mins RT
6. 3 wash tap water
7. incubation Sodium Thiosulfate 5% 5 mins RT
8. wash tap water and check |

osteologic disc but get weird results... Which cells are you using? MSC or
ESC or?

发帖数: 35 | 7 我也用了ALP， 结果都是negative的，好像protocol有问题，
所以现在做von kossa，好不容易有阳性了，结果都阳性。故此发问，
还请大家帮忙，这是我in vitro 的最后一个实验了，十分重要。 |
