mES cell line from it. Knowing that we could not isolate mES cell from
blastocyst in our facility, I tunred to iPS or nuclear transfer.

最近闲来无事整理了几个journal近二十年来的影响因子的统计(如图)
发现PNAS比较稳定,一直是10左右的水平,
nature/science 确实不相上下,缓慢稳步增长
不过cell和new england journal of medicine却在2000年左右有较大变化
2000年以前cell都是比NEJM高的
但是2000年以后NEJM越来越牛,cell却越来越不行了
2000年实现反超,从此差距越来越大
这背后有什么故事么?是因为cell有子刊的影响么?还是说NEJM有什么特别牛的策略?

请教版上有没有朋友做过细胞直径分布测量实验的,cell size distribution.
研究TSC-mTOR signaling 的经常要测量细胞直径,有人用流式细胞仪,也有不少用
cell counter的。
我最近做了几次,用的是cell counter,得到的细胞直径分布图一般都是不规则的曲线
,而文章里面的一般是类似正态分布曲线图
请问有这方面经验的版友,这个实验有什么关键的注意事项?

就实验研究手段而言,两者有啥区别?
satellite cell可以认为是muscle stem cell,我看到paper上大部分都是小鼠in vivo
的实验。primary myoblast实际上也是将satellite cell传代分离得到的。那么,假如
体外培养muscle stem cell,不就变成了myoblast?

不是很明白你这个问题。
传统说法是Pax7+ satellite cell变成myogenic cells, proliferate.. primary
myogenesis
then secondary myogenesis, myoblast cluster and form the myotubes.
体外养satellite cells的方式好像很局限。我还没看到好的protocl能体外扩增
satellite cell without losing the undifferentiate status.

t 细胞就是在血液,淋巴,组织只见循环, 血液和淋巴是最富集的地方,细胞回输通
过静脉,直接进入血液,也就直接接触血液癌细胞,因此不存在migration,
infiltration问题。
固体肿瘤会形成一个屏障,阻止t cell 的进入,同时固体肿瘤内部是一个非常强的免
疫抑制机制,进去的t cell 也很容易被抑制,也就是说,有限t cell 能够进入,进入
后功能又被抑制。因此在固体肿瘤上的效果不好。通过提高t cell 进入和抵抗肿瘤抑
制环境能力,就可以提高效果。
抗原当然也是一个原因,相比前面说的两个原因, 重要性底点,因为现在已经鉴定了
不少肿瘤抗原了。
cd19 car 是把几乎所有b 细胞都干掉,固体肿瘤里这样的抗原比较少,有也通常是组
织特异性的,会累计整个组织,器官。

比如 osteogenic cell line MC3T3-E1和 immortalized primary osteoblasts(自己
isolate and immortalize 的 osteoblasts)
要做一个实验,要拿cell line还有immortalized primary cell做对比,但我自己感觉
都是已经被immortalized的,就没什么区别了。值得试吗?
这两种细胞会有区别吗?谢谢啦

的杂志。cell metabolism差些。

Cell一直是想挣大钱的啊!Cell Reports不见得比Cell research IF高,至少再5-10年
之内应该不会发生。你随便看看Cell reports里面的文章的实时的citation就知道了,
相当不怎么样。这个和文章的质量无关。Nature好多文章都很bullshit, 但是不影响
citation. 这个是辩证关系和心理作用:同样的东西发在plos one上面就不如CNS上面
的citation高。大家都习惯cite high-profile journal papers.

我是发过cell research的,也帮老板审过该杂志的文章。
我的感觉这个杂志在审稿上还是比较专业的,尽管不排除有些文章编辑格外青睐,但是
绝大部分审稿过程还是很正式的。
发文章给钱这事楼主就不要去道听途说了!我是听都没听过。
再说了,你会为了区区3万块钱人民币,把你的一篇Cell投到Cell research上?都不说
cell了,NCB都不会有人干的。
有些事情用钱是能搞定的,但不是所有的事情。
每个杂志都有他们自己的几把刷子,要不咋在江湖混?

不过她们家的文章都很有意思,做神经行为方向,全是western blot 生化方向的。 很
少有组织学,生理学的实验。当然最后总会用一个行为学的实验来验证一下genetic
phenotypye,剩下的就是用生化解释pathway。感觉CELL老多都是这样的PAPER。起点都
是一个IN VIVO或者primary cell tissue, 中间做着做着,就看见293T cell, 然后
FLAG-TAG。我现在一看见有293T cell的文章,感觉都头大。那些生化PATHWAY用293T解
出来的,到底有多少是能真实的反应in vivo的生理。

照着Cell Stem Cell与Stell Cell Report的Editorial Board找就行了。stem cell的
牛人基本上都在这两个Board里了。
另外,干细胞,特别死胚胎干细胞与iPSC领域内的问题,很大一部分不是由于人为造假
,而是由于干细胞的特性决定的。与其他的细胞不一样,干细胞对enviroment的适应性
太差,很容易就会发生改变,而且很多时候,仅凭少数的评价指标(比如morphology,
marker gene expression等)根本就区分不出来。
但不可否认的是,干细胞确实是一个非常好的modeling platform;最近几年,生物与
基础医学领域的许多大的突破,都得益于干细胞的研究,比如epigenetics, NGS的应用
,translation medicine的发展,等等。

仅仅依靠一个两个所谓marker基因的相对表达水平来鉴定cell identity是阻碍干细胞
领域发展的最大障碍之一。
看上去很美好的Lineage tracing系统其实问题挺多,最大的问题就是干细胞往往非常
rare,很难detect到。而你detect到大量label成功的细胞不一定是真正发挥功能的干细
胞。
今后的希望还是回归到最本质的问题:what is cell? 细胞是执行生物功能的一个功能
单位,所以cell identity还是要从功能鉴定入手。所谓stem cell,不在于是否表达
marker,而在于在体内(或体外)是否有self-renewal的能力,是否能在分化过程中产
生出其他生物学功能。
关于功能检测方法多多,可操作性强一点的就是看看几个功能相关基因的表达。硬要讨
论“marker”的话,几个功能相关的基因协同表达倒可以算是个有意义的“marker set
”。

A 阳性的细胞只有total bone marrow的1%。in A positive cells, 30-50% is KSL
cells.
Transplantation assay shows that A positive cells are HSC enriched cells.
A ko mice are viable, but HSC function in A ko mice is not investigated yet.
I plan to generate a reporter mice, do you think will it be interesting?

对二流的期刊,业内评价就可以,但是对一流的期刊,业内评价有用吗?要更大的同行
评价才行。
期刊最终要转为经费才能衡量,生物界自己评价cell水平高,人家不给钱没用啊。各个
学科一起去金主争经费的时候,生物界认可cell牛逼有什么用啊?还不是要外行认可才
行。cell每年再多发一倍的文章,只能让cell的文章贬值,而不能让生物界的经费增加
一倍啊

it is very strange, not many solar cell companies have good R&D groups
my research realm is silicon heterojunction solar cell(not mid-bandgap part
in a tandem cell but a-si/c-si solar cell) and.... play with new device
structure and fast process
i knew United Solar/First Solar/Sunpower......
but there are very limited vacancies in their R&D groups.

a tandem cell with three junctions. The highest efficiency was achieved at 300
suns (AM1.5). It is certainly true that the built-in potential of inorganic

在做相关项目,上来胡说几句,当是抛砖引玉,大家多多指正
otto warburg老早就发现cancer更喜欢glycolysis,还因此得了诺奖,之前的肿瘤代谢
主要在体外造影上有一定的应用,为啥肿瘤代谢要到现在又火了呢。在这个field里真
正让代谢又火起来的个人看来是三个topic: PKM2, IDH1/2, Lkb1/mTor/AMPK。
尤其是Lewis Canteley 2008 Pkm2的nautre,让人们知道了cancer cell是如何建立
Warbug
effect的,这之后各种可能和warburg effect相关的pathway又被人们陆续挖掘了出来
