发帖数: 1 | 1 印象中大部分的载体上的CMV启动子组合都是CMV enhancer+CMV promoter，比如pEGFP-
C1载体。
最近实验室从SBI买了一个载体pCT-Tubulin-GFP，看了公司给我的map，我也测序验证
了下，看到这个载体上的启动子只有CMV promoter，没有enhancer。
这个载体使用以及基因表达完全没有问题。我就是突然好奇想向各位老师请教下这个
CMV enhancer是否是必须的？理论上CMV enhancer肯定是能增强CMV promoter的效率。
但是如果没有的话例如SBI这个载体用起来也没有啥问题，表达量也是刚刚的。哪位老
师可以给讲解下CMV enhancer的功能吗？有没有谁比较过with enhancer和without
enhancer的CMV promoter的基因表达效率? 谢谢啦！ |

做reporter。阳性的对照用的是CMV作为promoter，能看到GFP，但是效率非常。查了文
献，有的说CMV能在S2里表达（但量比较低） 有的说不能（那里说的是不能再KC细胞，
没有专门说S2）。所以想问下 有做过的 给个准确说法呢。 另外我要是就只想看看
有没有promoter activity的话，只用GFP作为reporter 而不是GFP跟其他蛋白的fusion
做reporter 是不是也可以的呢？ 新手呢，求狂拍 |

（CMV promoter，GFP tag），阳性对照用了另一个载体表达GFP看到很强的表达（另一

发帖数: 444 | 5 CMV promoter 在mESCs里面会沉默，对吧，比如mE14、J1，用CMV驱动表达是不行的。
但是我想用minimal MCMV promoter，前面接一个multimerized DNA binding sites，
后面表达mCherry，在ES细胞表达能结合DNA的转录因子，看mCherry。这个会有问题吗
？我的载体将会是
-DNA binding sites-minimal MCMV promoter-mCherry-PGK-puro-
如果不行。pGL4载体是用minimal Promoter，驱动luciferase，想问这个minimal
Promoter是啥？能用在mE14吗？可以用GFP换掉后面的luciferase吗，有这样的载体可
以分享／买的吗？最后得到一个这样萌萌哒的载体：
- DNA binding sites - minimal Promoter - mCherry - SV40 early enhancer/
promoter- hygro -
这个用在mE4没问题吧？

的seletion,然后挑克隆,虽然没有看到GFP的信号,但是我还是冒险试了一下EB的培养,
结果发现EB的表型和预期的一样,但是用WB检测的时候,看不到融合蛋白的表达.老板看
了不是很相信,非要做出wb,检测到蛋白,才能说明gene 的功能和你的EB表型的关系.我
搞的那个很郁闷呀, 构建, 转了8个gene呀,3个多月的工作呀,伤不起呀.
要是能检测到蛋白,这个project都差不多可以close了. |

1.CMV promoter可以drive shRNAmir的表达，见图1,干扰效果，CMV与H1和U6效果一样
。文章为
2005年nature genetics的Second-generation shRNA libraries covering the mouse
and human genomes.
2.Openbiosystem shRNAmir的设计采用相似的策略。CMV/tuboGFP/IRES/puro
