touch promotion due to previous reception of $100 promotion. Only one
promotion in one year.
我的背景是今年一月开的第一个account，上个月关的。关的时候并没有开始当前ipod touch promotion yet.
过程有点奇怪，具体如下：
（1）打上面客服电话，结果一听说是promotion的问题，就transfer到了我的local
branch!(难道上面的没有下面的懂得多？)
（2）local branch的人上来先问我哪里开的，我说网上，然后就把我的情形介绍了，
我主动说了previous promotion（反正是我的就是我的，不是我的不强求）。那个CSR
让我hold on了一会儿就说了，no。我让他then close my account，结果说因为没有

year review 的表格里有一项是自己对明年position 的expectation.
具体情况：
自己进公司的时候工资被打压了（因为年景不好， 公司当时基本不招人），后来的新
人们， ｔｉｔｌｅ ， 工资都比我高一级， 一年前（至此我已经工作了2年）才
ｐｒｏｍｏｔｅ 我和别人一样的ｔｉｔｌｅ 和工资（ 也就是说和和别人刚进公司
的ｔｉｔｌｅ 一样）， 但是去年比我晚进公司的一个同事已经又ｐｒｏｍｏｔｅ了
一级）, 我想提出明年要求promote一级（这样就和我前面提到的那个同事一样）， 是
不是应该在mid-year review 提出？另外怎么说比较好?(担心公司会以去年刚promote
过作为理由拒绝我， 但是我去年promote是因为刚进公司被打压的太厉害， promote
了也才赶上新人的待遇)。 另外上面不止一次说我的performance 是整个组里最好的，
但是这个待遇让我觉得不公平，之前太包子， 从来没提过任何要求。

发帖数: 425 | 48 做了个construct，转primary newborn astrocytes，用hygromycin来筛选stable
transfection，可是一直都没有筛出来，细胞全都死光光了。转染效率还算可以，是电
转，有30%左右。hygromycin浓度也不算高，50ug/ml。
hygromycin是用TK promoter表达的，不知道是不是因为promoter不行。试了别人的一
个puromycin construct，是pGK promoter，也基本上死光了。想问一问版上牛人有没
有经验可以传授。多谢多谢！
另外我是用CMV promoter来表达我要的基因，先转细胞，然后筛选stable line打到老
鼠上。听说CMV promoter在体内可能会被shut down，是不是也应该换掉呢？谢谢！ |

发帖数: 35261 | 49 我用玩具矿车打比方给你解释一下吧。转录就是小车(RNA Pol)在轨道(DNA template)
上滑行。你远远地站着往起始位置（promoter）上扔车，如果运气好车掉到轨道上了，
会有一个初始推力让小车沿轨道开始滑行。这就是转录起始。
原核生物转录起始比较简单粗暴。只要有promoter就能稳定转录。真核生物转录的要求
比较高。你扔小车的频率和小车掉到promoter上的概率都比原核中小。所以真核基因用
enhancer作为辅助recruit RNA Pol。1的情况就是不含enhancer的转录：可以转录，但
是水平很低。2.我觉得不是不行，毕竟原核也是用TATA BOX，只是位置比较固定。我猜
hsp之类的promoter如果位置合适可以一试。 |
