more data or do far more in-depth analysis prior to submitting your story
to top journals. Below are my specific questions:
Where do you obtain this repellent? From plants or from insects?
Can you elaborate on what ecological impacts this repellent has? Can it be
used as insecticide?
If you are sure this repellent is a mixture instead of a pure chemical, can
you identify its chemical components by doing HPLC, per say?

What genes have you identified by behavior-based genetic screen? Among these
genes, have you found any GPCR receptors or other transmembrane proteins?
If yes, you should focus on characterizing these genes as they are potential
repellent receptors.

Neuron ablation is one method to identify which neurons participate in
response to this repellent. However,
many drivers are dirty and you should narrow down which subset of neurons
are really recruited by doing more sophisticated genetic labeling
experiments. Moreover,
calcium imaging analysis is required to back up your anatomical mapping
experiments.
GPCR
If you want to try high-profile journals such as Neuron or CNS, identifying
the receptor is the key. ，

my boss is dreaming about sending this to a top journal now, so I'm curious
how other people think about it.
With current data I mentioned above I do not think it can go to top journals.

That's not my project anyway. We are not a neuro lab and my boss is doing it
herself.
The repellent is a social signal. The screen was done on genes known to be
involved in chemotaxis/olfaction in
this animal. No GPCR is tested because there are too many. However key
components of the GPCR pathway are involved.
I suggest my boss to identify the components of the repellent using HPLC and
do some calcium imaging instead of using laser ablation only. My boss says
that since we do not have time and expertise for those experiments we will
not do them... orz
I really think that without clarifying the chemical components or
identifying the specific receptor, it can go nowhere (from my point of view,
knowing it's a GPCR or knowing which G-alpha is involved sounds like
nothing)....... while my boss is
thinking about something like PLoS Biology...
