Major Project 1: Salivary gland-based gene therapy for lysosomal storage
diseases
Key achivements:
Fabry disease is a member of the family of lysosomal storage diseases, a
group of
monogenic disorders characterized by deficiency or lack of a class of
enzymes such as
apha-Galactosidase A in Fabry disease (GLA). Recent studies demonstrated
that an
intrinsic endocrine pathway is present in salivary glands and this distinct
pathway has
been shown to be capable of robust synthesis and secretion of on-regulated
gene products
into the vascular space. We generate the AAV vector containing the sequence
for human
GLA delivered to the salivary glands in alpha-Gal A deficient “Fabry” mice
. The specific
aims involve examination the organ biodistribution of the enzyme and
optimization of
dosing and immune physiology to achieve long-term expression of the enzyme.
Major Project 2: Adenovirus gene transfer of anti-biofilm peptides to the
salivary gland
Key Achivements:Gene transfer to the salivary glands presents the
opportunity to achieve
endogenous production and salivary secretion of peptide therapeutics.
Endogenous
production overcomes many of the challenges that have thus far limited the
utility of
emerging anti-biofilm peptide molecules for targeting individual bad actors
in the
periodontal flora. We generate the recombinant adenovirus containing the LL-
37
antimicrobial peptide and delivered to the salivary glands in rodent model.
The aim is to
achieve a steady-state, sustained concentration of the therapeutic in the
saliva and
evaluate the antio-biofilm potency of saliva from treated mice.
Major Project 3: ultrasound-assisted Aquaporin-1 (AQP1) gene therapy for
functional
restoration of salivary gland.
Key achivements: We have recently reported the development of an ultrasound-
assisted
gene transfer (UAGT) method in the salivary gland. This technique enables
transient,
non-invasive, non-viral gene transfer to the salivary gland using "off the
shelf" FDAapproved
microbubbles and clinical ultrasonography equipment. By using next
generation mini circle non-viral vectors, we will scale our UAGT technique
from mice to
swine, perform AQP1 gene therapy using UAGT in the salivary glands of rats
and swine
rendered xerostomic by ionizing radiation and characterize the proteome of
saliva
produced by treated animals with and withous partial sparing of the salivary
gland by
intensity-modulated radiation (IMRT).

Key Achivements: Invasion of bladder epithelial cells by uropathogenic
Escherichia coli
(UPEC) contributes to antibiotic-resistant and recurrent urinary tract
infections (UTIs),
but this process is incompletely understood. In our work, we provide
evidence that the
large guanosine triphosphatase dynamin2 and its partner, endothelial nitric
oxide (NO)
synthase (NOS [eNOS]), mediate bacterial entry.

Key Achivements: Dynamin2 is a large GTPase that regulates vesicle
trafficking, and the
GTPase activity of dynamin2 is required for the multistep process of
adenovirus infection.
Activity of dynamin2 may be regulated by post-translational phosphorylation
and Snitrosylation
modifications. In our study, we demonstrate a role for dynamin2
Snitrosylation
in adenovirus infection of epithelial cells. The results support the
hypothesis
that dynamin2 S-nitrosylation following eNOS activation facilitates
adenovirus infection
of host epithelial cells.

(WSSV)
Key Achivements: Apoptosis serves as an important defense strategy employed
by host
cells against viral invasion. Many viruses contain the anti-apoptotic genes
to block the
defense-by-death response of host cells. Using this marker rescue assay, we
identified
the function of ORF in WSSV is a kind of apoptotic suppressor like P35 in
AcMNPV.

Major Project 2: Inhibition of Measles virus multiplication in cell culture
by RNA
interference
Key Achivements: Measles takes a toll of one million people a year worldwide,
especially in developing countries and novel approaches to this problem are
needed. In
this study, we investigated the possibility of inhibition of Measles virus (
MV) infection
through RNA interference (RNAi), namely the interference with the second but
main
receptor of MV, signaling lymphocyte activation molecule (SLAM), in B95-8
cell line.
The results suggest that RNAi can silence SLAM expression and inhibit the MV
infection
of host cells.
