filled你的理解是个误区... filled也是表面一层，只是比plated厚。
filled是用金箔辗在base metal上，plated是用溶液做类似电镀那种。
filled印象中要求金层的重量要占到总重量1/20以上才允许叫gold filled.
“按一定比例参合在一起”的是K金。 |

RRS的板子最大的问题如图所示，板子的底部太厚，导致卡在arca-style的clamp上时
无法严丝合缝，有很大空袭，也增加了同一个clmap跟不同plate之间通用性匹配的难度。
好像这是RRS板子的普遍问题。
Lee speical 100mm push-on filter holder system.是专门为几个中大幅面的95mm口径
广角头设计。最大的好处就是不会有暗角和遮挡的问题。无须任何adpater ring
原来设计的目的是直接套在镜筒上。然后插filter，再在最外面加一个cpl
但是因为cpl会在最外侧，所以口径加大为105mm才能消除暗角，好处是方便调整cpl
但是105mm口径无论lpl还是cpl都价格不菲，并且这个口径的PL都没有coating，所以
我考虑是否可以用价格差不多的95mm口径有coating的CP |

kinds of plates. I am at a loss which one to buy.
I have a canon 60D camera, and a canon VIXIA HV30 camcorder. I'll purchase a
new camcorder soon. And in the future, I am planning to buy a Canon Mark
5DII. I hope the plate can work on these camera and camcorder.

做agar plate,没有那种灭菌工作台,就是在自己的bench上,点一个煤气灯. LB agar倒
上去后,盖上盖子,盖子一会就有很多蒸汽形成的水.我看有人是在灭菌工作台里面,开盖
子晾一会.在自己的bench上就不可能了.这个大家是怎么解决的?
还有就是含有抗生素的plate,加抗生素时候溶液不能太烫,不过也不能太凉,是不是还有
点烫手的时候加?大家是怎么掌握的?

That no colonies grew might result from many things. Not the right
antibiotic or not the right amount, or transformation went wrong... You need
to include at least a positive control and negative control each time when
you do transformation in order to troubleshoot.

about half the work. Tecan, Biotek, and BMGlabtech plate readers are all
better than MDs. I only used Tecan one but both Bioteck and BMGlabtech
based on their specs offer more function such as alpha scan. Tecan's liquid
handling is better though, if you are actually buying the liquid handling
portion. |

96-well plate，测生长曲线很有用。Tecan可以加一个staker，能够自动load几十个
plate，也很不错。Beckman的也不错，不过得换不同的filter比较麻烦，不过其biomek
FXp系统就很好用。 |

激发装置，荧光检测仪，以及合适的滤镜，随便什么荧光蛋白都可以检测。
但是，大部分plate reader，都没有装置这种设备，要么就是读透光度，要么就是
读自发荧光，这些装备是不能用来看激发性荧光的。 |

在做bacterial artificial chromosome constructs，实验室没人做过,大家都比较紧
张。我在筛clone,按照别人给的protocol,clone 长出来后，先挑在100微升的LB,长一
会，再分开几部分作functional test, pcr,之类的筛选。因为medium有tetracycline,
要避光，我就把含有colony的eppen tube放在盒子里，shaking,这样既避光，也方便。
然后，下午老板问我了这个问题，他说，老板娘告诉他，somebody is shaking plates
(其实他们知道是我，因为用的是30度 shaker,就我在用，他们有时要避光的plates,也是放在盒子里，在放在这个shaker,因为没有另外的30度incubator）
这样的常识我还是知道的。就算一时糊涂，也是浪费一些电而已。在这个实验室，我已
经是小心小心再小心了。知道老板娘，什么都看在眼里。可是你别看错啊，还马上打报告。真郁闷。 |

应该不是因为省电的问题，实验室再省也不会不开shaker, 那点电钱相对于
实验室材料的钱和你的工资来说简直就什么都不是。
我猜他们应该是担心你不懂干活乱做实验，把agar plate （？） 放进去摇。
稍微解释一下就好了。等你一些初步结果出来之后，他们知道你是能干活的人，
一般来说应该就会好一些。

皿。准备去别的实验室蹭microtiter plate reader，问题是OD超过1或者2是不是就要
稀释呢?问过他们实验室的人，也不清楚。请教了！
另外用nanodrop测DNA呢？上次测了个500ng/ul,应该也就不准了吧？

reader is the absorption assays. I have tried Bradford or OD280, both give
high standard deviation. However, if you just want a guess, try using your
sample at original conc, 1:1 dilution, and 1:4 dilution in triplicate. You'd
better have BSA standards at least 5 concentrations in the same plate in
triplicate. These will give you a relatively reliable readout. OD is linear
at 0.5 to 1, roughly. good luck |

问题是这样：
在倾斜混凝土面上立wide flange column，一个选择是浇个小的混凝土Pad，得到水平
面，然后放柱、base plate，浇grout；另一个说法是，不浇混凝土pad，直接浇grout
pad就行了，5 Star Non-thrink Grout。
问问大家，不用混凝土Pad可以吗？？在坡度上base plate以下需要厚度约为 1-6
inches。

As for those photoresist, you can get rid of them using O2 plasma etching.

快磨没了。今天去做BMW club的tech inspection，被告知得换前轮胎了。
换轮胎事小，但要是过一个月新轮胎又磨没了，那就太花钱了。跟technician讨论了一
下解决方案，似乎只能换第三方camber plate（stock plate的调教范围太小，没法帮
助），调negative camber（他说start with 2.5degree）。材料加人工一共要1，900。
还有technician建议换Hawk的brake pad，不如Pagit RS29那么hardcore。

5是个正确的解决方案. 但camber plate 不需要这么多钱, plate本身只有600，安装费
