2、sonication时，当时tube中裂解液的体积是不是很重要，大约多少ul比较好？
3、用液氮闪冻一下，是不是必须的？
4、PMSF要不要加？磷酸化酶抑制剂要加么？
5、用磁珠的话，也要preclear么？
