汽车年检没有过，读代码P0325,说是要换knock sensor。
去了一家修车铺，被告知：零部件很便宜，15美元就可以；但是部位是在发动机下面，手
工要好几个小时才能换好。手工报价是800-1000美元。
太贵了！！简直不敢相信。
回来后自己做功课，发现网络上有这么一个东西：

基本上是这样：
原来的部位不是非常难换吗？那把连线剪掉，然后连接到新买的knock sensor上面，然
后把它安装在intake manifold, which is just on top of engine。手工呢？只需要
20分钟，自己DIY都能搞定。换好之后，就能通过年检了。
但是，心里还是不放心：sensor还能够这么随便乾坤大挪移啊。
请问这里的大牛：自己把knock sensor这么挪位置，不会有问题吗？
如果没有问题，我打算就这么来了。

今天才确认， 我的车加速的时候，引擎有knocking声，
其实我一直都能听到这个声音，但不知道这就是 书里面说的 “knocking”
以前以为是exhaust系统的声音。
今天老美同学说 这个就是knocking
现在的问题是，
书里面说这个knocking是ignition方面的问题，
我的 spark plug和线都换过了，现在怀疑是 ignition coil的问题
但是又不确认，想买一个吧，担心换了以后不解决问题，（像以前的很多次一样）
所以，有没有个能试配件的地方？
想买个 used的， 如果能确定就是coil的问题，买个oem的也行
就担心又白花钱，
大家能说说么？

发帖数: 42 | 7 maxima的老毛病,DTC 0304就是knock sensor坏了,那sensor位置比较特别,但其实只要2分
钟就弄出来了.
器材:
12英寸的3/8延长杆(大致这个长度,长点的好)
3/8的弯头(英文叫flex, 就是能弯头的延长杆,不要软的,一定要硬的那种,自己到店里去
看)
3/8的ratchet,12mm的socket,这个就不说了
厚橡胶手套(医用的不行,要用厨房用的那种,当然你的皮厚就不用这个了)
方法:
1.等车冷了(不然皮就没了)
2.用延长杆, flex, 头上套12毫米的socket,伸到intake里面,套住knock sensor的螺丝,
稍拧松,然后拧1-2转(你在拧的话socket就顶到上面的intake了,这样连socket都拿不出来
)
3.拆掉intake旁边的EGR
solenoid的电线插头,如果你手比猩猩大的话,得把整个solenoid拆掉,一般只用拆插头就
行了
4.左手(注意!除非你不是人,右手就不要伸了,弄进去也没用的)戴好手套,把手伸进去,慢
慢用手拧松螺丝,取下sensor
5.取下sensor座上的门字形钢丝,取下旧的knock |

发帖数: 762 | 9 手动车。某一天不小心带速时候猛抬了一脚离合。于是熄火了。结果第二天发现在急加
速的时候发动机会哗啦哗啦响。根据本地车行的诊断这个是spark knock。并且我
youtube上的视频也完全符合这个症状。然后，我就加了一些cleaner到燃油里，又烧掉
了1箱premium的油。在这过程中check engine亮了，检查发现是混合气过稀。2个月前
换的氧传感器，一直都没问题。所以应该不是氧传感器的问题。是不是我那个时候抬离
合导致点火时机出问题了，要调整点火时机了？还是仅仅是那个时候刚好发生的，跟抬
离合没什么必然联系，换一下燃滤就可以了？由于我的spark knock在热车的时候非常
明显，是不是我也要考虑冷却水循环水泵有问题导致了引擎局部冷却不足？还有没有别
的可能性？上次花了100多才检查出来是spark knock。这次先自己诊断一下，如果实在
不行了再送去做engine diagnosis，估计还要上百。还不知道修好要多少钱。。。 |

knock sensor结构很简单,里面是一块圆环形黑色压电陶瓷,
两根线接出来,中间是一个固定用的铁套筒,螺栓拧在上面.
由于中间是铁套筒,所以压电陶瓷上都包有塑料绝缘薄膜,
以免短路.
这样的设计,用什么汽油都是震不碎的,即使你拆下来用榔头
敲,也很难敲碎.
knock sensor出问题主要是高温导致外面塑料壳开裂,造成
里面绝缘膜老化损坏短路. |

被他爸发现了x型腿，就是不能膝盖脚踝同时靠在一起，大部分时间膝盖靠在一起的时
候，小腿是往两边分开的。我俩一时间非常紧张，在约医生之前，google了一下，才知
道这个叫knocking knees。小孩子两岁到四岁之间多少都会有一点，是长身体的时候平
衡所需要的。一般来说，五岁之前会自己矫正，如果六岁以后还没有好，才需要担心。
我还看到一个俄罗斯的矫正成人knocking knees的网站，看起来几个月就可以把很严重
的给治愈。所以大家不用太紧张，小孩子有一点x型腿，不用太担心。

多谢答复，我所用lentivirus vector上都带有tubo GFP,所以puromycin筛选时,我拿到
是100%绿的cell line，对照组相同浓度的puromycin在一天内就可以将没转染的细胞杀
死。
我要knock down的基因是个转录因子，在发育中决定cell fate determination。
一个set中三个克隆都不起作用，很少见吗，以前做knock down比较少. |

要用小鼠做.做neuron knock down,你是往小鼠脑子里面打病毒么?需要cannula把.如果

我有一些很基本的问题想请教大家，
1，通常大家怎么标记老鼠，打耳洞，剪指甲？哪一种比较方便能够
追踪每一只老鼠？
2，大家应该用软件管理老鼠吧，一般那种软件比较常用？每只老鼠需要记录那些信息？
3，自己实验室养老鼠，大家还严格纪录代数吗？比如F1, F2, ....Fn等，F应该是兄妹
自交吧？如果回交的话，比如F3和F1杂交，那代数怎么算？还有如果两种Trangenic老鼠
杂交，比如Cre line跟flox line杂交，最终拿到flox homozygous的老鼠，这一般至少
两代，这每一代该怎么记录，这肯定不能算是F了。

发帖数: 31 | 35 最近很是郁闷，做啥都不顺！又碰上一问题，向大家请教。有一个和癌症相关的蛋白，
它的功能最近被发现能促进cell proliferation，这一现象被好几个group报到了并且
在不同的肿瘤细胞中都是相同的作用。他们用的方法都是使用siRNA knock-down这个基
因，然后用MTT assay 或者是thymidine incoporation assay去测cell proliferation
.我为了方便，就用了lentivirus (pLKO.1 based vector, open biosystem)做了
stable cell line,试了两个细胞系，knock-down 效果都不错，从western 上来看，
估计有80%-90%的reduction. 可是一测cell proliferation ,cell proliferation的抑
制效果都不明显，我用了两种方法：promega的MTS assay 和 BrdU incoporation
assay.这到底是怎么回事啊？我看了他们做的方法，最大的差别就是他们是transient

个exon前面，这样homozygous的老鼠该酶实际上是knock out的。
理论上GFP表达细胞是受该基因调控的，现在的结果是，在已知正常情况下该基因表达
的器官，homozygous的老鼠GFP细胞数大概是heterozygous的2倍多，比较的是相似位置
的切片，做过统计，基本上是这个结果。
请教这种现象准确原因是什么？Allelic Imbalance造成的吗？好像没什么理论基础支
持这个？另外，该基因也不存在明显的imprinting，应该不是这个原因吧。请高人指教

我们刚拿到一个knock in mice，刚刚用插入片段的引物做genotyping确定了germline
transmission.
关于如何keep这个line,见过一种观点，是最好一直用long range pcr来keep colony，
维持一个完整的选择压力，也就是说如果一直用插入片段的小片段pcr，可能会导致意
外地保留了一些随机事件，在某一代出现飘变
但是long range pcr成本当然要比普通pcr高多了，光酶就贵了n倍——
另一些常见做法就是就选择一个特意的插入片段，甚至neo casette来genotyping
在大多数实验室似乎也work得很好
不知是否有有经验的同学来谈谈？ |

Sorry for not making this clear. By saying knock in I am referring to the
method utilizing homology recombination to replace an original gene allele
by another one pre designed.
What you are saying is usually refereed as 'transgenic event'.
expression cassette will be randomly inserted into (any) genomic DNA.
can delete, mutate or add (tags etc) to any part of your allele in vitro,
then knock it in. |

，于是把exon1和exon2 replace掉（留下一小部分flanking sequence）
因为这个基因的3'UTR的调控做得很热，因此我就不能引入外源polyA 比如SV40, BGH
polyA等
粗看过去似乎问题不大，我用的EGFP的编码做过修改不含有splicing context
但是也有人说不加外源polyA的knock in表达很靠运气？ |

在下正在用siRNA knock down几个基因，使用的是电穿法（脂质法效果不好），所用
siRNA都是公司里已经validated的但有2个target gene却出现问题，A基因敲除后，
mRNA和蛋白都效果很好，但是control siRNA 却使靶基因表达量增高很多，这是怎么回
事啊？
B基因更离谱，siRNA 反而使靶基因增高表达3-4倍，这是脱靶效应吗？是siRNA 浓度太
高了吗？

For example, if you are studying a kinase function in neurobiology. You
found total KO of your gene leads to embryonic lethal phenotype, and
conditional KO of your gene in neuron leads to very interesting phenotype.
Culturing of KO neuron in vitro also showed dramatic phenotype.
Interestingly, restoration of kinase dead (KD) mutant in KO neuron fully
rescued the phenotype (same as restoration of WT).
