dos.img镜像用ultraISO写入USB闪存盘，即制作好了启动盘。
把marker目录及文件copy到制作好了的U盘启动盘上。
设置bios, 用U盘启动.
◆DOS下 进入marker目录
CD marker
◆①命令bak.bat备份BIOS到BIOS.ROM文件
bak.bat
◆②[然后，用16进制编辑器查找 E3EDC4000100010003000000 ，如图修改BIOS, 只需
改2个字节就可以了]
注：
现在Thinkpad的任何BIOS其实都是含有SLP 2.1信息的。（有SLIC 2.1表的，就不多说
了。不需要任何操作，直接装OEM win7就是了。）
DOS机没有SLIC 2.1表的是这次的修改对象。
大家备份一下Bios, 得到BIOS.ROM. 用16进制编辑器(例如Windows下UltraEdit)打开
BIOS.ROM，查找十六进制数据块十六进制E3EDC4000100010003000000定位。

。但并没有很多stem cell 的 in vivo marker被identified出来。比如说liver
tissue,都知道其可以再生，但是它有没有stem cell仍然在争议。更莫提marker了。
endothelium是个快速turn
over的组织，但起在成体内stem cell population的identification仍然未知。
筛选stem cell marker的方式可以有两个途径
第一就是正向的从已知marker出发，比如说通过已知marker筛选出某stem cell
population, 分析其有啥gene 高表达，以此出发建立CreERT transgenic model来确认
新的marker.
对stem cell来说找到marker不是目的，更重要的是这个maker背后的mechanism. 一个
新的marker市时常可以揭示新的regulation mechanism.

做一个实验，需要co-stain cell surface marker 和intracellular protein,然后拍
照。细胞是adherent 的。
因为担心fixation和permeabilization会影响cell surface marker,所以准备先活细胞
染 cell surface marker，然后在fix，permeabilize染intracellular protein.
我的问题是可以活细胞染 cell surface marker， 马上fix，然后再染 2nd antibody
anti-cell surface marker吗？
还是必须活细胞染 cell surface marker，再染2nd antibody anti-cell surface
marker， 完了才能fix？

上次讲过入门paintball最好的marker 就是A5+flatline barrel。 这marker射程远，
皮实，性价比又高， 继续推荐. 基本价位最好的面具还是强烈推荐JT flex 8
Thermal. 非常舒服,防雾，保护又非常全面。tank买68／4500 HPA. 几个大牌子都不错
。 我用的是Ninja Carbon fiber. 纯个人喜好，没所谓。 如果不想$100+的话说，可
以买48／30。黑猩猩的只要37。 性价比最高。 就是差不多每俩个game(500 balls)就
要refill.
如果同学也开始喜欢paintball， 开始上瘾的话。 A5可能就不能提供需要的性能。例
如如果玩speedball, 就会被其他player out gunned. 大家都ramping, marker又小又
灵活。

得到的结果优于少数“精心”挑选出来的marker
并不能说明那少数marker没有价值。
老夫曾经由于其它问题研究过在混合高斯模型下，
从高维数据中随机抽取部分特征后的“表达力”问题。
发现这随机抽取的特征只需要满足很弱的协相关
性的假设，那么这些特征的表达能力跟用全部特
征差别不大。具体取决于全部特征本身的表达能
力，以及抽取的特征数量占全部特征数量的比例。
由于这样随机抽取100 个marker以后，其表达能
力跟用全部基因的表达能力差别不大，所以这100
个marker的表达能力就很强了。因此它们会跟“精心”
挑出来的少数基因结果差不多甚至更优越。
当然话说回来，目前的很多相关研究都是有问题
的。自欺欺人地在用统计却不管任何条件和假设，

可行。
lineage tracing不完美，比如leakage等问题。不过已经是当前能做到的最好的了。依
然该是金标准。
没有一个marker是完美的。比如alexandra joyner的那篇关于GLi1+ cell作为hair
follicle stem cell marker的文章。单纯从lineage tracing结果来看其实并不完美，
其对hair follicle的contribution并不完全，但是重要的是他们揭示了这群gli1+
cell是受神经分泌的Shh的调控的。这就重要了。marker不是目的，找到一个marker就
意味着一个in vivo model的建立以及一个可能的新的regulatory mechanism.
到底什么是stem cell.我很赞同你的观点。。stem cell 到底是个identity还是个功能
？ 这个问题现在很难回答。 |

发帖数: 1007 | 26 找到一个marker后能开展研究的方向很多，上下游只是一方面。
除此以外，基于这个CreERT model研究其表达pattern ,Stem cell dynamic本身就很有
意义了。比如说Hans Clevers组做的Lgr5, Lgr6在肠道里的那几篇Nature, Science文
章，就只是研究其表达位置，还有拿confetti reporter来研究其激活后populate gut
epithelium后的方式。对其上下游signal pathway基本没咋研究。
就像你说的，其实很多干细胞marker真的只是凑巧在那里表达，没啥重要的功能意义，
比如NG2，nestin，俩都是最早是在brain里面被拿来标记神经干细胞的，然后做MSC的
发现其标记MSC不错。但这俩的knockout并不会导致啥MSC的功能缺陷。还有Gli1也是。
其功能对于MSC来说很弱，有点像是凑巧在干细胞那里表达。
干细胞的marker 可以是功能性的，比如ESC里的sox2, oct4啥的。也可以是非功能性的

发帖数: 130 | 27 仅仅依靠一个两个所谓marker基因的相对表达水平来鉴定cell identity是阻碍干细胞
领域发展的最大障碍之一。
看上去很美好的Lineage tracing系统其实问题挺多，最大的问题就是干细胞往往非常
rare,很难detect到。而你detect到大量label成功的细胞不一定是真正发挥功能的干细
胞。
今后的希望还是回归到最本质的问题：what is cell? 细胞是执行生物功能的一个功能
单位，所以cell identity还是要从功能鉴定入手。所谓stem cell,不在于是否表达
marker，而在于在体内（或体外）是否有self-renewal的能力，是否能在分化过程中产
生出其他生物学功能。
关于功能检测方法多多，可操作性强一点的就是看看几个功能相关基因的表达。硬要讨
论“marker”的话，几个功能相关的基因协同表达倒可以算是个有意义的“marker set
”。 |

erase marker, wet erase marker, and washable crayon etc.

GFP+arteriole marker. ephrinB2 staining如果过太复杂或者涉及antigen retrieval
,GFP可能会受影响。
另外我找到一个smoothelin 文献说特异表达在arteriole的SMC里，而不在venule的SMC
.
形态学上动脉和静脉还是很好区分的。但是我想最好还是用多个marker以及形态学一起
来验证我的GFP-expressing cell的位置。所以才在寻找这些marker |

作者抓了几十篇paper里面报道的所谓在breast cancer里面表达有区别的基因，和他随
便抓的基因比较，只要抓到100个基因以上，两个结果就相差不显著了。
然后他看了一下发表的那些所谓marker，大部分都和cell cycle/PCNA有关，如果拿
PCNA的表达做normalization，世界清静了，cancer marker基本失效。
结论就是cancer marker就是看细胞分裂的，没啥奇怪。 |

SA-b-Gal方便，但是假阳性挺高。在培养细胞中用起来挺方便，看组织切片就困难多了。
SAHF，rH2A.X，53BP1也会有DNA Damage的假阳性。
Ki67，BrdU是negative marker，更不好说了。
谁要是找到一个specific的marker就好了 |
