1. 10-2003 确诊，07-2004终于手术切除，期间Jobs一直在试自己选的diet疗法。扫描发现癌变在扩大才动了手术，这段时间癌转移的多少不可知，但肯定越拖转移的越多。
2.手术后没化疗和放疗。Jobs 2005年演讲说这表明病情乐观，不知是哪个庸医给他灌输的。2004年手术是大面积切除，部分胰腺和胃和小肠，胆和胆管都切除，目的是要消除扩散，但不系统化疗和放疗怎么能最大程度减少扩散威胁？哪个外科大夫这么信自己的手艺？
3.2009年换肝，这在扩散性癌治疗是有争议的，因为换肝后压制免疫反应有可能促进别处癌的生长。也有可能当时肝已经不行了没办法。
这种癌正常治疗大多数可活10年以上，Jobs真该到东岸Hopkins或Harvard找个靠谱的大夫。

Negative，不需radiation。
biospy时左乳发现的两个tumor：
乳头处的tumor HER2是negative (IHC score 2+，FISH negative) ；
12点的tumor HER2是positive (IHC score 3+)。
手术病理显示有两块 specimen, 因为有re-excision。
但是只发现一个 tumor(乳头处)，1.5cm, HER2结果和biopsy时乳头处的tumor一样，
IHC score 2+, FISH negative，另一个(12点)不仅tumor没找到，biopsy时放到那的
clip也找不到。后来作过X-RAY， US， MRI，没有发现体内有什么残留。

最近在做一个project。和一个治tumor的药有关系。在一篇文献里，用的是BDF1 雌老
鼠和E0771 solid tumor。 24mg/kg 剂量的95%纯（有约5% 的一个impurity)的药 注射
。14天后average tumor size 只有control的2%，但21天后tumor size 长到control
的21%。也就是说虽然药有效果，tumor有会回复并没有完全控制。
在稍后的另一篇文献里，药纯度99.5%。用的是human MX-1 mammary carsinoma 在nude
mice。 3 mg/kg的99.5%的药注射。14天后average tumor size 只有control 的0.3%
。 21天后tumor没有回复。文献的作者说这结果很好，剂量低了10倍药效反而好，是因
为开始5%的impurity给出去的结果，因为这个impurity会有competitive binding。
我们现在对这个药很感兴趣。想知道是不是药API纯化后药效真的变好很多。不知道这

以在合适的环境下生存，条件适宜的话会发展成转移的肿瘤。
术后化疗就是为了尽可能杀死这些潜伏的癌细胞。化疗的fractional kill性质，一次
不能杀死全部癌细胞，重复的疗程能尽可能接近全部杀死癌细胞。
这就是其实你母亲的决定是有道理的。清除越干净，复发几率越小，间隔时间越长。
化疗的副作用，换来的其实是更长的生存时间。
一些已经转移的dormant tumor cells是无法被杀死的。他们会静静潜伏多年，在适当

Malignant tumor equals to cancer. Benign tumor is very common, and usually
is not fatal unless the tumor grows in unusual places. For example, benign
brain tumors could be fatal. This is because the space within skull is very
limited. When the benign tumor grows bigger and bigger, it will push other
brain parts. So its not the tumor itself kills patient but the "compression
effect" of the tumor causes the trouble. |

1. "If cancer strictly follows evolution model, should one imagine certain
genetic lesions gain dominance?" 某些肿瘤可能的确是这样的,比如一些EGFR突变
的NSCLC,表现出oncogene addictive,这种genetic lesion似乎dominate肿瘤的生长,因
为对突变的EGFR进行target therapy（erlotinib）具有疗效．当然oncogene
addictive也并不排斥CSC.
2. 关于癌基因会诱导正常细胞senescence or apoptosis.这是事实，但一种可能的解
释是，这种或这些癌基因并非是tumor initiator,它们只是promoter或maintainer;也
就是说在它们异常之前，有其他genetic lesions作为tumor initiating factors，而
这些我们还没找到？

1. Control cell with geneA
2. Control cell knockout geneA
3. Tumor cell with geneA
4. Tumor cell knockout geneA
1~2 样本是配对的，3~4样本是配对的。第二张图的连线情况就是配对情况
PCA 的图和基因表达分析吻合的有两点：
1. control 和 tumor 基因表达pattern大不相同，基因分析结果也吻合：比较这两者
时有上千的基因显著的up or down regulated （比较红色和蓝色）
2. PCA 显示Normal sample中有无GeneA，基因表达变化不大。这个和array分析结果也
吻合。（比较红方块和红小球）
但有一点PCA 的图和基因表达分析不吻合：
1. PCA 显示 在tumor中有没有GeneA，表达pattern也应该有相当显著的区别，但我用
fold change>3; FDR<0.05的阈值算，只有58个基因有显著变化。（比较蓝方块和蓝小球）
