别的。现在一般笔记本的power adaptor只有65W，如果你idle就有30w，那一运行笔记
本的power adaptor不烧掉了？
现在一般笔记本都是至少500GB硬盘了吧，和这个Acer一般大。
笔记本组要是方便，提起来就走，搬家啥的都不怕。台式机再小也不方便啊。 |

prefered network, 头都大了. 将电脑refresh到出厂设置还是不行. 只是最近几个星
期的事,以前都好. 另外不可能是router的问题,因为我另外的一个电脑一点事都没有.
我的电脑是HP DESKTOP, 用的wireless adaptor. Windows 8.

我是把 dest.server:139 ssh tunnel 到 windows loop adaptor 的 IP 上的，好像文
件共享不受影响，因为 windows loop adaptor 和 localhost 的 IP 不同。
至于想 ssh forward samba，是想尽量在 firewall 上少开洞，而且这样所有的攻击都
可以在 sshd 上看到。
嗯，让我查查比较现成的 script/program 来自动识别攻击并封锁对方的 IP。过去的学
校的 IT 很强，所以自己不用太操心，server 被攻击后不久 IT 就能发现并且处理。现
在这个学校的 IT 就是一团糟，什么都得自己操心，呵呵。

发帖数: 697 | 12 哈哈，搞定了。原来是DVI Adaptor 上面一根针歪了没接上。其实早注意到了，只因为歪的形状和位置特别好，看起来似乎是制造的时候特意让那根针不连接，就没有怀疑。查了所有DVI Adaptor的格式，没有这一种，才怀疑是针歪了。一掰过来就一切OK。估计是换Monitor的时候不小心把它弄歪了。

故欢乐Monitor （都是Dell的LCD, 但Size相差很大）。就没有任何信号了。如果把
Monitor接在主板的VGA接口上，无法Boot，因为“Your computer has an add-in
video card." 如果接在add-in video card 的DVI （Dual DVI Splitter) 上，听声音
似乎Boot了，但Monitor 看不到信号。大侠们请帮忙。 |

号设置中 只有调制解调器的连接，点击 ”建立连接” 后显示可选择要建立连接的选
项有“电话线已与调制解调器连接”或手动设置或通过局域网…这样的选项。但没有发
现 Netgear USB Wireless Adaptor。 请问如何设置才能让IE 利用已经配置好的
Netgear USB Wireless 连接呢？ 难道必须把系统升级到Win2000以上？(老电脑好像不

差较明显；另一个是tripod adaptor不行，装上脚架一碰就晃得不行。
我觉得要买高倍望远镜，至少tripod adaptor不要是那种一根小棍拧上的，而要那种能
固定在镜筒上的架子。另外还要一副好的三脚架。

用RNA Ligase加一个自己设计的adaptor（DNA)到RNA的3'端，
然后用gene specific forward primer和adaptor specific reverse primer扩增
5'RACE也可以用这个方法做
我试过这个方法的3' RACE，还蛮方便的
因为我做的原核，没有poly-A tail，没有kit可以用
这个方法帮了我大忙

很大程度上这一步的效率取决于这个VTTTT-anchor primer跟poly-A tail的结合程度
我个人觉得既然你现在做不出来
很有可能是cDNA synthesis这一步就已经效率很低
我当时做RLM-3’RACE的一个原因就是原核没有poly-A tail
没有办法用poly-T的primer来做cDNA synthesis这一步
RLM-3'RACE带来的好处是
mRMA3'连上adaptor以后
后面的cDNA synthesis用的是adaptor specific oligomer
这个效率会有不同
如果已经山穷水尽，这个结果又一定要做出来
可以试试这个RLM-RACE的方法
不过就是多设计个引物，买点T4 RNA ligase而已
Roche的反转录酶还是不错的

另外如果你担心是因为mRNA的二级结构有影响的话
在把RNA加入cDNA synthsis反应之前
可以95度加热RNA3分钟，然后立刻放在冰上，冷却以后再用 |

NEB的更干净一些，直接降解含U的第二链。有专门的文章比较过不同的方法。
KAPA的貌似跟illumina类似。
如果买NEB的adaptor要注意他们卖的是loop adaptor，序列中有一个U，连接完了之后
要用USER把loop打开成Y字形才能扩增。
不如直接买NEB整套kit省心。 |

2. 路由器起个什么用？是不是如果通过电脑来共享的话就不需要路由器了？如果决定
用路由器，一定需要DD-WR或者Open Wrt吗？
3. 以后电脑就必须一直开着？我倒是有多余的laptop但是感觉电脑一直开着能行吗？
网上算了一下, Laptop开24小时每年的电费大概在$55块钱左右。其实是比直接买
premium的plan还是便宜了很多。 |
