如果8小时还没有附着的话,细胞肯定死了
不知道你的floating cells多不多,占多大比例?如果数目很少的话western,ladder根本
无法解决问题
我目前也在做这方面的研究,我通常使用flow cytometer来检测subG1或caspase,
但也需要有一定数量的细胞,fibrobalst实际上很容易培养的,你研究中出现的现象我也
遇到过,认为很可能是trypsin处理过头了,如果附着细胞还有不少,你可以把浮游的去
掉,继续培养剩下的细胞。

请问如何鉴定这些floating的细胞仅仅是floating而没有dying,还是它们已经进入cell

I am working on extracting total RNA from few PFA-fixed cells (about 200
cells).
Now we have MagMAX™-96 Total RNA Isolation Kit (Cat:AM1830). This kit
is for RNA extraction from normal cells.
Did anyone have experience on it?

Yes new iGluR is selective Silencer because it is selective for K. They
took so much trouble to get the selectivity right, and it is a great
improvement.
If ChR2 is selective for Na but not for Ca or Mg it will be much better.
Ca do so much more than depolarization. If we want to ultimately
introduce the channel into the brain to stimulate some neuronal cell, it
is better it won't cause Ca overload.

p53RE-Luc,这个luciferase是衡量p53和luciferase的什么参数
Caspase 3/7,这个量是衡量什么用的。
hERG, (VC): 20% inhibition @ 6 uM这个是什么意思啊。
这个是一个p53抗癌药物的cell assay,从专利上看来的。不是学生物的,google了一
圈,也看不太明白。
求大牛给个比较简单的解释,或者给个网站,偶去好好科普一下。

f the cancer cell line can initiate tumor, but is there a sub-population in
these cell lines that acts like CSC?
As for what is important, in clinical? It's hard to tell. Say, cell origin o

ring to the tumor-inititating cells identified in immune-deficient mice) ass
ociated with relapse, drug resistance and metastasis.
Can anyone point out an original research paper with some experimental data
showing these cells have any of the three major capabilities? My impression

I did the E coli (SURE2) transformation in the cell culture hood... as i
remember I did not drop the bacteria cells out, just some LB media...
I spray the hood with 70% ethanol for many times, and turn the UV light for
10min,
is it possible for the bacteria cells to contaminate the laminar flow hood???
What should I do to de-contaminate the hood?
help me! I do not want to get this trouble...

现在做的一个cell一用抗生素就碎掉了,本身又不太attach,所以
没法通过反复pbs洗来去除死细胞。我试了一个密度梯度离心histopaq,
有很多细胞被扔掉了,而且有点担心会不会弄死细胞。各位有什么高招?
我想用cell strainer把cell留下,碎片流掉。但是现在的strainer
多是40/70/100um的,有没有比较小的strainer或者mesh, 大概5-10um的?

No perfect methods. It largely depends on your research purpose.
Cells carrying beads are bad for their trafficking in vivo. But magnetic beads enrichment is quick and relatively economic, as compared to sorting. For in vitro study like RNA purification or culture, beads purification is fine.
Sorting is expensive and labor-intensive (I suppose your lab do not have
your own sorter and do not sort cell frequently). But without beads binding
, cells are more healthy.

I transfected different oncogenes into 293 cells. I'm wondering whether
some oncogenes may affect cells and make them sensitive to freeze down. If
so, there must be some factors changed by the oncogene. Then this should a
part of function of oncogene.

领域里面一个比较牛校的实验室,很多年的工作都是基于一个他们实验室才有的cell
line,他们没有描述这个cell line是怎么来的,只是一句话很笼统地说是从ATCC的细
胞subclone来的,引用文献是最早ATCC的那个细胞的文献。
我搜索了一下他们用的细胞的名字,所有的文献都是他们实验室的。给PI写了一封信,
问能否把cell寄给我。
但是我还是觉得,这样的工作,有什么意义?那个细胞又没有什么经济价值,到底是啥
不清不楚,发这种别人都没有办法重复的文章,有什么意义?

Washed once with PBS before adding fixation solution (3.7% formaldehyde/PBS).
Cells detached in fixastion solution. Is it because formaldehyde does not
work with HEK293 cells? Anyone tried methenol? What's the concentration?

I do use 45um filter to filter through the lysate before preparing the rest
procedures. I guess you can collect smaller aggregates less than certain
diameters.
The other thing is the purity of your cells. If the lysate is prepared from
heterogeneous population, where cells have different sizes, than you have to
be careful to isolate large cells and small aggregates.

I didn't read them again, but in my memory, these two papers tried to
address the same question: why chemotherapy drug,tamoxifen, suppressed
breast cancer cell growth, but promotes endometrial tumorigenesis. Shang's
paper showed that tamoxifen could trans-activate PAX2 gene expression in
endometrial cancer cells, and the activation of PAX2 resulted in the

我有好几年没怎么看MSC的文章了,但好像这些年这领域也没什么突破性进展。我就说
几句,供参考。
直接用bone marrow MNC和分离培养MSC再注射的文章都有很多。直接用MNC的考虑大概
是骨髓中到底什么细胞是真正的能够transdifferentiation的stem cell没有公认的结
论,有人说是是在MSC群体里,有人说是是在HSC群体里。所以从应用的角度,直接用
MNC而不是分离纯化好的细胞,不会遗漏任何可能的stem cell。还有一点考虑,全骨髓
移植已经成功应用到临床有50多年了,其可能的风险和经验已经积累了很多,现在只是
将其用于新的疾病,便于被批准。而MSC之类的,需要经过体外培养,近十年才开始小
规模临床试验,风险和效果未知,推广起来挺难。当然,这并不妨碍实验研究,MNC和
MSC都可以发文章。
关于是否用辐射,辐射只能损伤快速增殖的组织,比如骨髓,肝脏,小肠粘膜等,对血
管平滑肌损伤效果未必好。很多组织特异性的损伤都有其自己的方法,比如心血管用缺
血再灌注,胰岛用STZ。我不清楚血管平滑肌是否有特异性的损伤方法,但放射损伤我
不认为它是一个很广泛的适应症。

3-NPA does that. 3-NPA directly inhibits succinate dehydrogenase activity.
At sub-toxic doses cells are still viable, but SDH activity could be significantly inhibited.
MTT/MTS/Calcein Am/Resazurin assays are often over-used or somewhat used
incorrectly to measure cell viability/cell number.

As mentioned above, a lot of the cytotoxicity/cell viability assays
are based on mitochondrial function, particularly SDH activity. This renders
a lot of the assays under question when the "manipulation/treatment" have
potential impacts on mitochondrial function.

For cancer cell lines, it is less of an issue because of the
notorious "warburg" effect through which cancer cells are more glycolytic
and rely much less on mitochondrial OXPHOS.

做了好几年的工作,virus infection and host immune system的领域,攒出来一大堆
date,故事也说圆了,老板也认为不错,觉得我得工作是目前关于这个问题最完整最有说服力的研究工作,可惜老板一直觉得新意不够。
刚投了immunity,审稿一个月后,一个reviewer说很好没有任何意见,另一个说没有新意,结果editor以”没有新意“为由给毫不留情的据了。
和老板商量了准备换个杂志投,老板还是觉得novelty是个问题,不反对试别的好杂志但他个人建议赶快投个容易接受的杂志。可是俺实在不忍心看着这么多工作好几年的心血就去投个小
杂志。所以还想试试好一点的杂志,找了找,觉得cell host and microbe 和 PloS
Biology从IF上看都还很不错的。不过不知道这两个杂志是不是都很难投啊?有知道的
大侠给说说吧?例如是不是对novelty要求很高?
另外,如果immunity据了以后,投同是cell press的cell host and microbe 杂志,
editor是不是会知道原来投immunity被据的事?那是不是会很吃亏啊?
