GMI１０７０， Tbc1269， 基本都买不到，估计都申请专利了，因为selectin
inhibitor 有临床价值。
想问问版上的同学，有哪位知道如何能得到selectin inhibitor 呢
或者有什么selectin inhibitor 能够买到的吗
我查到R&D有一个pselectin inhibitor, KF 38789。 我想要E-selectin, L-selectin
and p-selectin inhibitor.

发帖数: 75 | 3 各位大侠们，我在做用6ohda处理细胞的时候，发现一个蛋白的水平降低，文献表明可
能是proteasome（实验是过表达此蛋白，然后用MG132处理，发现蛋白增多；）或者
caspase降解（在细胞中过表达此蛋白和各种caspase酶,发现蛋白降解）。
我于是用proteasome inhibitor MG132与6OHDA 同时处理，并没有发现此蛋白水平回复
（保证MG132其作用，有阳性对照蛋白）。换用caspase inhibitor和6OHDA处理，也没
有发现次蛋白水平回复，换用lysosome inhibitor和6ohda同时处理，也没有发现次蛋
白水平恢复。
请教别人，说proteasome的inhibitor MG132 并不足以阻止proteasome。说一些好的杂
志都不接受MG132作为proteasome的inhibitor。是这样么、？那有什么好的推荐么？
另外一种可能行是我用药物处理之后，蛋白水平下调，是因为我的蛋白沉淀了，在我的
buffer里面是没有的。但是我是用cell signling公司的buffer20mM Tris， 150mM 氯

lypholized powder溶解后aliquot出来很多小管，store在-20，用的时候拿出来放4C不
放回去，
这样容易坏吗？

hydroquinone monomethyl ether做inhibitor。只用一种单体的时候不除inhibitor都
可以聚合，可是共聚合死活不行。请问这个与inhibitor有关系么？谢谢各位达人～～

我正在学习step2,说出来大家讨论一下，不是说要用不同的机制的药物来控制吗，这明

想IC50 都是用in vitro 方法测得的, 真要加在细胞上用多少浓度呢? 应该和IC50 一
样吗? 看了一些文章, 好象treat 细胞的浓度都比IC50 大. 可大了又会有specificity
的问题. 大家都是如何解决的?

构的， 也就是说最好一个小分子bind Pol或者
initiation complex以后就抑制它们bind DNA的。大家有什么推荐么？

基本的意思是说， EXEL是做KI的， KI的成功率大概是４０％左右， EXEL的pipeline
上面大概有11个KI， 如果按统计规律， 应该有5个最后会被FDA
批准。
