最主要的是As妨碍普通细胞的呼吸oxidative phosphorylation，普通细胞没法在As下生存，不可能繁殖
但是这种几年前就被发现了的细胞有酶可以中和As，还可以靠As呼吸， 所以可以靠As生长，所以在用As替换P的环境下繁殖，就丝毫不足为奇了
所以更有意义的发现不是As替代P， 而是细菌可以在As里繁殖。这是个老消息了，可怜NASA不要脸，又玩花样骗钱
早就说过了，NASA是个很可怜的机构，在资本主义社会中没有任何存在的意义，所以要向好莱坞学习，不停搞出新闻来，才可以吸引缴税人民的注意力，搞研究经费

Of course the most direct and wildly accepted is to measure mitochondrial
respiration through a Clark-type electrode.
Alternatively, cellular oxygen consumption can be measured through an oxygen
sensitive probe (used to be called A65N, but now commercialized as
MitoXpress via Luxcell). This method only requires a fluorescent plate
reader in addtion to mitoXpress, which should be available to most
biomedical labs/departments.

来做临床诊断的才几十个，在模式生物里能用的才十几个。这个要想用老鼠来做，特别
是要做到genetics这个level，metabolism这种日节律很强的phenotype在老鼠上作超烧
钱是一定的。
或者，就直接screen病例，这样不用知道全貌，找到一个新的metabolite，或者一个新
的基因控制已知的metabolite，就算paper发得不高，也是很有用的啊。 |

FACS analyze adipocyte progenitor and do some functional assays
这种实验没有必要先做的。可能是放到你整个实验的最后那一步。
先是chow diet，检测体重，饮食量，ITT， ogtt or igtt, 血液指标，脂肪含量/体重
，有变化，做个脂肪组织和肝组织的切片看看。有变化再做个能量代谢。
如果chow diet，你的这个ko 小鼠都有变化，那就神了，说明这个基因很强啊。
不管有没有变化，用个High fat diet试试，也许能看出更明显的效果，重复同样的实
验。
如果想做的fancy一点，cross到ob/ob or db/db mice，这个要看你ko mice是什么背景了.
不要用什么STZ之类的。
最后做各种实验，分析机理。
B6j是做metabolism公认的，6n背景，有的reviewer会不认可，当然了B6j是非常容易诱
导成metabolism dysfunction的小鼠了。 |

发帖数: 37 | 45 最近一直在考虑career的问题，估计是年纪大了，想安定了。
个人背景：国内三流医学院毕业，国内生物硕士，加拿大生物硕士，毕业后留实验室做
技术员一年多。
目前考虑一年内办好移民后换份工作。个人没什么太大的追求，只想有份稳定、收入说
得过去的工作。考执照做医生是基本不去想了，不知道还有没有其它的选择。
以前读书的时候一直忙于实验（老板比较push),从未认真考虑过这个问题。
技术方面主要做分子和细胞的实验，目前管理10多个mouse lines,可以做MCAO,brain
injection这样的实验，也可以做切片、染色以及in situ。所做课题和metabolism相关
，GTT,ITT以及分离各种老鼠组织都非常熟练。不知是否有机会去一些公司做
metabolism相关的研究？
另外考虑过读个physician assistant的学位去医院工作，似乎更稳定点。
