我不知道这一段DNA是如何进入细胞核的，进去了，一直处于游离状态，人家细胞自己
的DNA开始转录，可外来的DNA自己又没有promoter，怎么开始转录成RNA啊？
另外一个问题，是不是转染到细胞里，双链线性DNA，环状DNA，都可以？ 那单链DNA可以吗？
还有一个问题，我们平时摇菌，扩质粒，这些质粒都自己可以独立复制的吧，都是基本不整合到细菌基因组的吧。我们摇菌用的细菌自己有自己的质粒对不对？

Most plasmid for transient expression will carry their own promoter upstream
of the expression codon.
DNA可以吗？
single strand DNA is degraded easily and practically not ideal for
transfection.
本不整合到细菌基因组的吧。我们摇菌用的细菌自己有自己的质粒对不对？
Yes. Most of them have their own replication origin site.
If you look at the vector map you will notice things like "F1 Ori".

using.
Most of them should carry a promoter. If not, then they won't get expression
in transient transfection. However, if they got integrated into chromosome,
they will.
I don't know how you get these questions, because they are not "real
questions".
It all depends on how you design the DNA for transfection therefore there is
NO real answers to it.

发帖数: 366 | 5 双链线形DNA，最大的可能是被细胞内的外切酶降解掉，也可能整合到基因组里。

The only thing they do is to provide a template for RNA transcription
because they all contain strong promoter such as CMV.
Becaue they don't replicate, their level will decrease and dispear after a
few days, that's why they are called "transient expression".
At low frequency, they may intergrate into genome, then they can replicate
along with the genome. You can use antibobitic to select this low frequent
clones, that is called "stable clone se |

expressed at high level for over one weeks (until I tossed them). They are
pretty stable in cells, is not that easy to be degraded (DNA or protein). It
's easy to test that, just cut off the ORF and transfect it. |
