好像也没有人去细究这些。其实mRNA 3' 端结合很多蛋白来调控mRNA的稳定性，保持不
被降解，从而在转录水平不变的情况下，mRNA水平上升。大家说说如果mRNA水平高，还
有哪些可能性造成？
另外就是如何设计实验区分这两种可能性呢？ |

发帖数: 1005 | 4 run-on可以说 Pol II富集和活性比较高，跟 pol II的Ser2状态一致，跟Ser5关系不太密切 ,与transcription stability没直接的关系吧？

发帖数: 363 | 5 normally people use rRNA (like 18S or 28S) as a control, so run on is
general for RNA polymerase but not only for Pol II.
Run-on reflects how many active polymerases were binding to the gene-of-
interest at the moment the cells were freezed. So it can reflect the
transcription activity (I did not stay that it can tell the stability). And
of course, you know nothing is 100% in biology, right?
nuclear run-on, just google it, easy to find. It is not good for short genes
, because less space for polymerases' binding there.
太密切 ,与transcription stability没直接的关系吧？

发帖数: 3419 | 6 谢谢各位。这个nuclear run-on不错，检测transcription rate，而不是像qPCR那样只
检测transcript expression level。我在想反应时间应该很重要，时间长了，比如6个
小时，mRNA肯定游走出细胞核，mRNA的稳定性也会影响信号强度，最后的结果就不好解
释了。反应时间短的话，10分钟，信号会弱的看不到，实验难度就会太大。谁了解mRNA
转录，转移，降解的大致时间点？谁有这个实验的protocol？ |
