i*********0 发帖数: 915 | 1 cell signaling ab for p-stat3. two bands.
what is the meaning of the lower bands? isoforms? or modification? |
n***w 发帖数: 2405 | 2 Is this a pY705 STAT3?
It may be recognizing both serine and tyrosine phosphorylated stat3. |
D******9 发帖数: 2665 | 3 could be a small isoform. there are many reports about small isoforms of
Stat family. however, some researchers think they are caused by protein
degradation during cell lysate preparation. |
i*********0 发帖数: 915 | 4 I am using Phospho-Stat3 (Tyr705) (#9145) from cell signaling.
I favor the answer of small isoforms. Maybe they have different functions.
Thanks a lot, guys, for the answer. |

i*********0 发帖数: 915 | 6 Just got the reply from the CST. It seems the lower band is isoform beta.
Dear @@@@,
Thank you for your inquiry regarding our Phospho-Stat3 (Tyr705) (D3A7) XP
174; Rabbit mAb #9145. The top band, at 86kDa, represents Phospho-
Stat3alpha. The bottom band, at 79kDa, represents Phospho-Stat3beta isoform
. |
g*********9 发帖数: 3528 | 7 It is clearly stated on its website......
See background:
"Stat3 isoform expression appears to reflect biological function as the
relative expression levels of Stat3α (86 kDa) and Stat3β (79 kDa) depend
on cell type, ligand exposure, or cell maturation stage (10). It is notable
that Stat3β lacks the serine phosphorylation site within the carboxy-
terminal transcriptional activation domain (8)." |
