g******c 发帖数: 18 | 1 It's totally normal that such DNA bands are present on a same Urea-Page. The
band shape could be "smile"- or "cry"-like on the same lane. The reason
lies in the
ionic strength of the loaded sample. The closer the band is to the loading
well, the bigger skewing effect the sample ionic strength makes on the band
shape.
20 years ago, most labs did sequencing in house using Urea-PAGE. Most
seasoned biochemist witnessed such mixed shapes on the same PAGE.
The younger generation scientists may not be familiar with Urea-PAGE.
Ironically, a veteran biochemist also fell down to the trap today. I feel so
sorry for his post on twitter.
