你相当于问,如果我合成一堆塑料,陶瓷,加上一堆金属,堆在一起就能形成发动机么?
又相当于问,如果我有一堆元器件,他们随机堆在一起就能形成计算机么?
细胞并不是简单的生物大分子集合,细胞是一个复杂的分子机器,其中大量生物分子并
不是像溶液中一样自由反应的,他们被精密得组合在不同的时空中,在特定的地点特定
的时间发挥特定的作用; 这部分子机器精致到可以自己复制自己。
这样的机器随机形成是非常非常小概率的事件。你可以用下面这个例子想像一下它有多
困难:
你用一种计算机语言,C++,Java,python,等,把里面的有效字符,函数当成一个元素,
然后让他们随机组合在一起,企图自动产生一个可以在操作系统中自己复制自己的程序

一直在误导,现在还在误导。 说说明corona virus 不可以从蝙蝠直接传染给人,必须
经过一个中间宿体性的其它动物才能传染给人。所以,病毒发生后,就故意误导全国人
民去海鲜市场找证据,找其它野生动物的麻烦。 而且还是病毒所去找的,找完了还装
模做样化验呀,分离呀什么的。最后把责任全部推给了海鲜市场的动物。可是那种动物
,一直不敢说,说了其它相关已经在人就会去早那种动物监测。 所以压根不说,打马
虎眼。
事实上,早在2013 年,就是这个武汉病毒研究所,已经从来自云南的蝙蝠身上所携带
的corona virus中分离出第一株蝙蝠SARS类似样的冠状病毒的活病毒,其中就包含了类

现在红遍天下的光遗传学,大家(包括我)都认为是Stanford的Deisseroth创造的。但
是昨天我不小心看到了一篇新闻文章,才惊讶的发现其实最早做出来的,有可能是
Wayne State University的潘卓华。虽然他的投稿时间和Deisseroth的类似,但是因为
他是一个没有什么名气的学者,而且他的文章包装得不对,所以他的文章被百般耽误,
最后比Deisseroth的晚了一年才发表。
在2004年底潘就做出了和Deisseroth几乎一样的文章,除了用的细胞略微不同:他做的
神经细胞是从视网膜分离的,而Deisseroth 做的神经细胞是从大脑分离的。然后他把
文章陆续投向Nature, Nature Neuroscience,Journal of Neuroscience,却统统遭到
了拒稿。然后大半年之后,也就是2005年8月份,Deisseroth 的文章发表在Nature
Neuroscience上。潘得知之后,心都凉了。

This is a kind of pattern matching problem ?
My thought is:
1. First transfer all molecule structures into a string and store them.
2. Then search for the strings for the desired string.

Ft, do you know the principle of x-ray diffraction? It requires the molecule
to be arranged in an orderly repeating pattern. So how you gonna arrange
this pattern by single molecule? The protein solution will have molecules
randomly arranged to all orientations, x-ray technique is not able to do
anything with it, it needs crystals.

FRET is the best way to confirm interaction, although FRET between more than 2 molecules can be very tricky.

I won't agree with RY on drug development
Merck spends ~$8 billion on R&D each year (marketing and sales spending is a
lot larger than R&D), and it can get 2 new molecule drug approved each year
if lucky.
In China, 一类新药 doesn't mean new molecule drug. China never developed any new molecule drug.

I forget the source of information. It may be scientifically misleading, so
use this information at your own risk.
For GFP molecules in mammalian cells, the common ground is about 1000 GFP
molecules. Somebody ever claimed that 400 molecules were good enough.

Again, if A binds B strong, you can try native PAGE with increasing amount A
from zero to whatever maximum, the resolution of native gel might be
sufficient for you to distinguish B/Ai from B/A(i-1). If indeed, you will
get n+1 band on the native PAGE when combining all lanes: B, BA1, ... BAi,..

Single molecule quenching might work as well. Label A with GFP with near 100

其实如果我们换个方法脑洞一下,纯属假设,不要在我的帖子后面瞎回啊,我没有想替
老韩翻案的意思。
现在大家(包括我在内)都认为NgAgo用老韩的材料方法做不出来。
但是假设真菌体系里确实work了,而且不是在大于37度的条件下做出来的,那说明
NgAgo 在真菌里不需要高温也能起作用,而且说明NgAgo有可能需要assistantance
molecule才能起作用。而这个molecule很有可能是真菌里才有的,考虑到老韩实验室条
件简陋而且石家庄雾霾大,有没有可能他做出来的几次(假设他真看到了)是碰到了不
是特别严重的真菌污染?这样转细胞时顺便把真菌给弄死了,而且神奇的
assistantance molecule也一并进入细胞帮助了NgAgo的编辑?
最后再说一遍纯属娱乐,大家看着乐呵就行,不要把对老韩的不满发到我身上,嘿嘿

Anyway, R&D in Pharma/Biotech is very tough nowadays. Now big pharma has
troubling developing small molecules, and layoff a lot of people working on
small molecules. Later, if they are not doing well on large molecules, I do
not know what they need to do next.
