今天早上1点才住下，睡了4个多小时后，小龙和小龙女接我去race start，之后见到了
湾区诸位神仙Frank, Larry, cives,
julian, feline, paff, Le. 比赛开始第一迈就是一超大上坡，跑起来和走路速度差不
多，旁边的人基本都是走上去的，我也连
走带跑。上坡延续了40多分钟。Larry在我出生的时候就已经BQ，现在专门训练trail
ultra，上坡下坡非常自如。我跟他一
起跑了40分钟之后就决定不能继续跟他跑，不然一定crash。之后不久cives就追了上来
。他今天以MP effort跑30K，聊了
一会儿之后cives往前冲，很快就没影了。这样上上下下，我跑到半马距离时已经花了1
：55多。brief bathroom break之
后来第二圈。第二次跑这几迈的大上坡已经非常痛苦，而且这还是有了每周一次15％
hill run的底子，否则会非常狼狈。
跑到坡顶时我感到很绝望，觉得前半程跑快了，甚至考虑把50K改成marathon。原本今

Took two falls, my first time on the trail. One was quite major and got a bleeding had. It's a "fat ass" race, we paid only $25 and got well stocked aid station every 5 miles. But I was a bit surprised that the Race organizer didn't even have a first aid kit. I had to use water and salt to disinfect my wound, talking about masochistic ultrarunners...

这是我的第二次全马，成绩是3:08.37(unofficial), 比今年的SFM首马快了大约25分钟
(其中至少5分钟是因为course的原因)。SFM首马没有什么目标，有些打酱油的感觉，为
以后PR做了很好的铺垫。这次因为要想BQ，所以全力以赴，以后想PR每分钟都会很难了。
这次准备CIM，我跟的是AM55mpw12周的计划。准备SFM时比较无知，没跟任何计划。后
来读跑版知道了AM宝典，CIM的训练就决定跟它了，从AM的5周recovery跟起。一边跟计
划一边读书，对其中的训练慢慢增加了了解。计划一直到11月初都跟得很顺利，中间还
参加了3个半马的比赛。Norcal 半马(1:34)让我觉得BQ有戏，SJ RnR (1:29)又让我有
了新的目标，MHM半马只按MP心律跑的结果(1:35)使我对实现新目标有了信心。但一切
在第八周的10k Tune-up后就都改变了。那天正好比较冷（50F而已，只是我第一次在中
午<60F的温度下跑 tune-up, 而且是第一次跑10K），即使热身了2迈多小腿肌肉还是有

没有跑过其他race.
25号的前2周的2次tempo都感觉不好。根据jack Daniels' formula + 2个月前的5k成绩
，我应该跑57:14分钟（<9:15pace)。但是9：05的tempo(4mile而已）都跑的不舒服，
担心达不到。所以给自己定的goal A 是57:14分钟， goal B 是1小时以下。
礼拜六我常发呆，ld问想什么呢，我：＂strategy.＂， 他：＂。。。＂
当晚做恶梦，跑了最后一名，大家给我打气，63分钟完成。
礼拜天，突然天就又变回冬天了。
一开始先压着9：15 pace跑。每到一个mile marker 听人喊时间来check.
一路都特别轻松，简直就像平时跑easy short/long, 和跑5k race的感觉完全不同。过
了4mile 以后我就没刻意check速度了，怎么舒服怎么跑。
不知为何，我一直以为10k是6.3m. 我的习惯是还有0.2m时开始提速，还有0.1m的时候
冲。事实证明我搞错了,最后有点措手不及。

她race，我鼓励女儿去和那个小男生在草地上race，心想，女儿灭了他。女儿估计不知
道是累了还嫌人家长的不够帅，拒绝之。哈哈哈。。我家女儿就是有个性，不鸟他。

啊，于是和他画下道道，从操场这头到那头在回来。我心想，咱虽然年纪大了，但是无

Marathons in the mountains of eastern Tennessee completed the race, the
first time since 2007 that the endurance test had no finishers.

In 30 years, 14 out of about 1,100 runners have completed the race, made up
of five loops around a mountainous 20-mile course. With a finisher rate of
about 1 percent, the Barkley has been labeled by many as the world’s
hardest race.

发帖数: 680 | 12 free play里只有车的出厂调教吧？那个当然不行。
里面所有的车都有（很大的）调教上升的空间。买上全套racing的悬挂、变速、sway
bar、笼子，前后定风翼（我不用racing 轮胎因为太容易就突破class限制了），然后
用网上很流行的ForzaTune计算器算一个基本的悬挂配置出来，你会发现和出厂配置差
别很大。然后你可以试跑一下你很熟悉的赛道看看效果，一般就差不多了，如果有轻微
的over或者Understeer再微调一下前后的sway bar就好了。那个计算器调出来效果还是
不错，唯一让我觉得不舒服的是他的悬挂总是偏软，不是都说跑车悬挂比较硬的么。
如果是高速赛道比如那几个Oval的或者Le Mans，可以做一套拆掉前后定风翼的调教，
这样不会损失很多最高速度。其他弯道多的，尤其是有高速弯角的，lateral G的一点
点差别就可以差很多。
如果你喜欢玩漂移模式的，更是一定要做调教。调教了以后更容易让尾巴飘起来。
要是跑Drag Race，变速箱一定要调
要是从来没摸过后驱车的话，建议一上来不要碰B class以上的车（这个级别以上的都

more challenging. Only for 3' RACE, i bought all the reagents from different
companies and combine to do it and it is cheaper. For 5' RACE, it is very
important to get full length mRNA or cDNA. Some kits can help you to achieve

CDS全长2K多一右，想知道转录起始和终止位点，于是做5/3RACE，5'RACE很容易就做出
来了，3'RACE就怎么都没有结果，3'端很多AT，尝试了降低延伸温度到60－65度，还是
没有结果，不知道如何是好，求指点！ |

很大程度上这一步的效率取决于这个VTTTT-anchor primer跟poly-A tail的结合程度
我个人觉得既然你现在做不出来
很有可能是cDNA synthesis这一步就已经效率很低
我当时做RLM-3’RACE的一个原因就是原核没有poly-A tail
没有办法用poly-T的primer来做cDNA synthesis这一步
RLM-3'RACE带来的好处是
mRMA3'连上adaptor以后
后面的cDNA synthesis用的是adaptor specific oligomer
这个效率会有不同
如果已经山穷水尽，这个结果又一定要做出来
可以试试这个RLM-RACE的方法
不过就是多设计个引物，买点T4 RNA ligase而已
Roche的反转录酶还是不错的

另外如果你担心是因为mRNA的二级结构有影响的话
在把RNA加入cDNA synthsis反应之前
可以95度加热RNA3分钟，然后立刻放在冰上，冷却以后再用 |
