you can let your faith affect your judgment and interpretation of the
evidences, but it is already a fact that chromosome 2 is widely accepted as
the end to end fusion of two chromosomes

when you recover telomere, you are immortalizing a cell, so the cell high
much higher chance to become cancer cell. If you can recover telomere of a
whole person, I would guess this person become a big big tumor.
Why? some cell should die after a few passages of division. If they don't
die, the balance will be broken.
so, elongate/recover telomere itself greatly raise cancer risk.

决定人的寿命的两大因素:
一,人只要活着就有新的细胞产生和老的细胞死去,产生新细胞由染色体复
制,然后细胞分裂,但是细胞不能无休止地分裂,细胞也有寿命。每个细胞的染色体上
有个“telomere”,中文叫端粒,每分裂一次就短一些,直到到终端,细胞的寿命就终
结,这也是人为什么终有一天会死亡,科学家已经开始研制如何延长telomere。
二,超氧化物 “superoxide”对细胞的破坏,形成细胞变异、死亡。人在消
耗能量的过程中主要采取有氧分解,在这个过程中,如果过多的氢原子,但是氧原子不
足,就会产生大量的超氧化物(H2O2),所以很多人说心宽者年轻,这也不无道理的。
人着急、生气的时候会感到胸闷,这也是肺部缺氧的一个征兆,所以经常着急、生气的
人会多产生超氧化物,就会容易短寿和变老;心宽者凡事大度,心胸开阔就会增进肺部

关于二号染色体的问题很多,尚无定论,远远没有证明或者弄得像是人和黑猩猩同祖先。
比如说,如果是融合的话,理想的证明是在二号染色体上找到两个着丝粒(
centromeres,在染色体的中部),但是,Ken Miller所引用的那篇文章说:one of
the two centromeres became inactivated (2q21, which corresponds to the
centromere from chimp chromosome 13) and the centromeric structure quickly
deterioriated(LaDeana W. Hillier et al, Nature 434, 2005),换句话说,只找

这个好像挺常识的了吧,我不是做这个的但是seminar和看文献提到的还是挺多的
刚去随便搜了ncbi的review,这个摘要里就讲了“. Telomere dysfunction has been
implicated in aging and senescence, and shorter leukocyte telomere length (
LTL) has been demonstrated to predict cardiovascular disease and mortality. ”
至于是不是primary defects, 不是特别明白你指的是什么。据我所知只是确证了端粒
随着细胞分裂次数而缩短,而端粒太短与很多疾病相关现象有关,如触发凋亡或者染色
体融合,这个应该是有非常solid的实验证据的

你说的恢复机制之类的,可能是指telomerase吧。生殖细胞有这个表达用来对付
telomere erosion.
细胞生存过程中,除了分裂的时候有telomere erosion问题,还有别的DNA damage问题
。生殖细胞里也不一定可以恢复,但是可以依靠自然选择(精子竞争游泳,有问题的受
精卵流产之类)。

1 北京大学生命科学学院魏文胜课题组
我们使用了文章中报道的高效ssDNA,同时自己设计了其他2条针对体内其他基因位点的
ssDNA,与NgAgo表达质粒共转HEK293T细胞与HeLa细胞,2天及5天后,用T7E1检测切割
效率,这三条针对2个不同基因的ssDNA,在HEK293T和HeLa两个细胞系中,无论转染一
次还是在8到12小时后再转染一次ssDNA,都没有检测到切割。
针对敲除后能使细胞有表型变化的某特定基因,我们设计了ssDNA,与NgAgo表达质粒共
转HeLa细胞5天后,发现细胞表现出了一定的对应表型,但是把这部分细胞收集起来提
取基因组,对靶位点测序,没有发现基因组序列的任何改变。
我们针对某特定基因设计了6种ssDNA,用特异的荧光报告系统检测是否有靶点DSB产生
,结果显示所有实验组均没有检测到对靶位点DNA序列的切割活性。

我们在293T细胞中表达带有Flag标签的NgAgo ,在不同时间点检测NgAgo蛋白表达。发

本千老告诉你,人老死的原因是器官衰竭,器官衰竭的原因跟电脑CPU用的太久挂了的
原因是一样的,磨损了。人和电脑不一样的地方是人的细胞可以繁殖复制,但是有些器
官比如心和脑子,结构太复杂,需要在发育的时候一次成型,类似倒模,倒模成型了就
不能替换里面的部件了,除非彻底把它拆了,所以就算单个细胞可以繁殖更新也没有用。
只要解决器官替换,人就可以无穷无尽的活下去。
但是你最后会遇到泰修斯之船的哲学问题,一艘船,所有部件全都换过,还是同一艘船
吗?人体同理。

日本精神科医师渡部芳德在其书《一个月,从癌症生还》中记述,自己在一次健检中发
现肺癌。他下定决心,开始透过饮食、健走,以及运用正念来调养自己的身体。一个月
后他再回医院做检查,原本3~5公分的肿瘤竟消失无踪!
渡部芳德在书中讲述了自创的“抗癌(想像)健走法”。身为精神科医师,渡部芳德非
常了解情绪与疾病的关系:当人处在焦虑状态下,免疫力就会下降。于是他很快收拾自
己负面的心情,采取一种积极的“焦虑控管”技巧:在每日健走的时候,集中心力对抗
癌症。

Older fathers have uglier children, researchers have claimed after linking
age to genetic mutations.

Not exactly true. For body building, a high protein diet is usually recommen
ded. However, a high protein diet is not healthy for the kidney in the long
run and easy to trigger inflammation. Multiple studies have shown that calor
ie restricted mice live longer than mice with regular diet.
I personally still believe that human body is like a machine with limited se
lf-repair function. As cells divide by themselves (maybe after 50 passages?

前一阵JAMA上有篇文章说鱼油还可以防止telomere缩短,间接延长寿命。
多吃点没错。
