anyway，要做dot blot的话不推荐用pvdf，用nc的比较方便
另外如果有vacuum blotter那做出来就很漂亮了

发帖数: 301 | 4 (1). Spot your cell lysate (or culture supernatant) onto nitrocellulose
membrane. 2-5ul is good enough for each spot. Positive and negative control
is a must.
(2). Let it sit on the bench to dry. 20 minutes is more than enough.
(3). Block with 5% milk or BSA at room temp. 30 minutes. Gentle shaking.
(4). Add your primary antibody (let's say mouse monoclonal antibody) to the
milk directly. Room temperature 2 hours (or cold room overnight). Gentle shaking.
(5). Wash three time with 1XPBS. 15 minutes/wash. Gentle shaking.
(6). Replace your wash with AP-conjugated 2nd antibody (AP-anti-mouse
antibody) in 1XPBS. Room temperature, 2 hours. Gentle shaking.
(7). Wash as described in step (5). Gentle shaking.
(8). Add one-step NBP/BCIP to cover the blot. Positive dot should show up in
5 minutes. Gentle shaking.
(9). After dots show up, dump NBP/BCIP solution and rinse blot with water.

时间这么短会不会影响结果质量？难道一般的protocol都是浪费时间？

ｂｌｏｃｋｉｎｇ过夜，一抗过夜。但是如果你就等一个western结果，很多地方是可
以加快的。

曾几何时，转膜4度过夜，一抗4度过夜，第三天看结果。
